Description	This AAV used in the gene knockdown experiments as a control in cultured cells and animal experiments. The 1.2 Kb EF1alpha promoter is a commonly used, constitutive promoter derived from human elongation factor-1 alpha (EF1alpha). It is often used in conditions where the other constitutive promoters, such as CMV, have diminished activity or have been silenced. EGFP is a basic (constitutively fluorescent) green fluorescent protein published in 1996, derived from Aequorea victoria, with a Excitation/Emission spectrum around 488nm/507nm. It is reported to be a rapidly-maturing weak dimer with moderate acid sensitivity. The regular plasmid miR30-based shRNA knockdown vector system is a highly efficient method for transiently knocking down expression of target gene(s) in a wide variety of mammalian cell types. Unlike standard shRNA systems, which utilize RNA polymerase III promoters such as U6, miR30-based shRNAs can be transcribed by diverse RNA polymerase II promoters. This also enables the use of tissue-specific or inducible promoters.
