Alcian Blue Periodic acid Schiff/AB-PAS Stain Kit

Periodic acid–Schiff (PAS) is a staining method used to detect polysaccharides such as glycogen, and mucosubstances such as glycoproteins, glycolipids and mucins in tissues.
The Alcian blue molecule is a large conjugated dye molecule that contains a centrally located copper atom bound to a pthalocyanine ring and four isothiouronium groups. The isothiouronium groups impart an overall positive charge to the molecule. The cationic isothiouronium groups likely bind via electrostatic interactions to the anionic sulfate and carboxylate groups located within the carbohydrate moieties of acidic mucins. Neutral mucins that contain carbons with adjacent hydroxyl (1,2-glycol) groups are oxidized by periodic acid. The resultant dialdehyde groups react with the Schiff reagent.
Acidic mucins are stained with the Alcian Blue technique while neutral mucins and glycogen are stained by the PAS reaction.
