Interleukin 11 (IL-11) is a protein that in humans is encoded by the IL11 gene. The protein encoded by this gene is a member of the gp130 family of cytokines. This gene is mapped to 19q13.42. It is a key regulator of multiple events in hematopoiesis, most notably the stimulation of megakaryocyte maturation. IL-11 has been demonstrated to improve platelet recovery after chemotherapy-induced thrombocytopenia, induce acute phase proteins, modulate antigen-antibody responses, participate in the regulation of bone cell proliferation and differentiation and could be use as a therapeutic for osteoporosis. What's more, IL-11 can stimulate the growth of certain lymphocytes and it has functions in many other tissues, including the brain, gut, testis and bone. In transgenic mice, overexpression of the human IL11 gene resulted in the stimulation of bone formation to increase cortical thickness and strength of long bones. Functional note: Cytokine that stimulates the proliferation of hematopoietic stem cells and megakaryocyte progenitor cells and induces megakaryocyte maturation resulting in increased platelet production. Also promotes the proliferation of hepatocytes in response to liver damage. Binding to its receptor formed by IL6ST and either IL11RA1 or IL11RA2 activates a signaling cascade that promotes cell proliferation. Signaling leads to the activation of intracellular protein kinases and the phosphorylation of STAT3. Reported localization: Secreted Expression/tissue context: Heart. The fetal heart shows a greater expression in the atrium than in the ventricle, while the adult heart shows a greater expression in the ventricle than in the atrium. Isoform 6 predominates in normal adult heart. Isoforms 1, 7 and 8 are expressed in fetal heart. Isoform 7 is also expressed in failing adult heart.

Research relevance and current trends

Adaptive Immunity: Researchers commonly examine how Il11 relates to this theme using model systems and orthogonal readouts.

Angiogenesis: Researchers commonly examine how Il11 relates to this theme using model systems and orthogonal readouts.

B Cells: Researchers commonly examine how Il11 relates to this theme using model systems and orthogonal readouts.

Common research applications

Western blotting: compare relative Il11 levels across conditions; band patterns may reflect isoforms and processing.

ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

Specificity notes: No cross reactivity with other proteins.

Cross-reactivity: No cross-reactivity with other proteins.

Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.

Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

//cdn.shopify.com/s/files/1/0949/7424/7277/files/a01160-1-il11-primary-antibodies-wb-testing-1.jpg?v=1772608312
