KLK1 (KALLIKREIN 1), also called KLKR, is a protein that in humans is encoded by the KLK1 gene. KLK1 is a member of the peptidase S1 family. KLK1 is a serine protease that generates Lys-bradykinin by specific proteolysis of kininogen-1. The KLK1 gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19 and its exact cytogenetic location is 19q13.33. The KLK1 gene contains 5 coding exons. And KLK1 is the most centromeric gene in the cluster. Mice lacking tissue kallikrein are unable to generate significant levels of kinins in most tissues and develop cardiovascular abnormalities early in adulthood despite normal blood pressure. The protein is functionally conserved in its capacity to release the vasoactive peptide, Lys-bradykinin, from low molecular weight kininogen. Functional note: Glandular kallikreins cleave Met-Lys and Arg-Ser bonds in kininogen to release Lys-bradykinin. Reported localization: alpha-beta T cell receptor complex; external side of plasma membrane; immunological synapse; cell body; cell-cell junction; dendritic spine; integral component of membrane Expression/tissue context: Isoform 2 is expressed in pancreas, salivary glands, kidney, colon, prostate gland, testis, spleen and the colon adenocarcinoma cell line T84.

Research relevance and current trends

Angiogenesis: Researchers commonly examine how KLK1 relates to this theme using model systems and orthogonal readouts.

Cancer: Researchers commonly examine how KLK1 relates to this theme using model systems and orthogonal readouts.

Cardiovascular: Researchers commonly examine how KLK1 relates to this theme using model systems and orthogonal readouts.

Common research applications

Western blotting: compare relative KLK1 levels across conditions; band patterns may reflect isoforms and processing.

ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

Specificity notes: No cross reactivity with other proteins.

Cross-reactivity: No cross-reactivity with other proteins.

Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.

Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

//cdn.shopify.com/s/files/1/0949/7424/7277/files/A02789-Kallikrein_1_-primary-antibodies-WB-testing-1.jpg?v=1772608357
