Mediator of DNA damage checkpoint protein 1 is a 2080 amino acid long protein that in humans is encoded by the MDC1 gene. The protein encoded by this gene contains an N-terminal forkhead domain, two BRCA1 C-terminal (BRCT) motifs and a central domain with 13 repetitions of an approximately 41-amino acid sequence. The encoded protein is required to activate the intra-S phase and G2/M phase cell cycle checkpoints in response to DNA damage. This nuclear protein interacts with phosphorylated histone H2AX near sites of DNA double-strand breaks through its BRCT motifs, and facilitates recruitment of the ATM kinase and meiotic recombination 11 protein complex to DNA damage foci. Functional note: Required for checkpoint mediated cell cycle arrest in response to DNA damage within both the S phase and G2/M phases of the cell cycle. May serve as a scaffold for the recruitment of DNA repair and signal transduction proteins to discrete foci of DNA damage marked by 'Ser-139' phosphorylation of histone H2AFX. Also required for downstream events subsequent to the recruitment of these proteins. These include phosphorylation and activation of the ATM, CHEK1 and CHEK2 kinases, and stabilization of TP53 and apoptosis. ATM and CHEK2 may also be activated independently by a parallel pathway mediated by TP53BP1. Reported localization: Nucleus. Expression/tissue context: Highly expressed in testis.

Research relevance and current trends

DNA/RNA: Researchers commonly examine how MDC1 relates to this theme using model systems and orthogonal readouts.

DNA Damage & Repair: Researchers commonly examine how MDC1 relates to this theme using model systems and orthogonal readouts.

DNA Damage Response: Researchers commonly examine how MDC1 relates to this theme using model systems and orthogonal readouts.

Common research applications

IHC/IHC-F: assess spatial distribution of MDC1 across tissue regions and cell types using matched controls.

IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.

Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

Specificity notes: No cross reactivity with other proteins.

Cross-reactivity: No cross-reactivity with other proteins.

Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.

Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

//cdn.shopify.com/s/files/1/0949/7424/7277/files/A01252-2-MDC1-primary-antibodies-IHC-testing-1.jpg?v=1772608354
