PDCD1 (Programmed cell death 1), also called PD1, encodes a cell surface receptor that is a member of the B7 superfamily involved in immunomodulation. This gene is mapped to 2q37.3. PDCD1 acts as an inhibitory molecule on T cells after interacting with its ligands PDL1 and PDL2. The PDCD1 gene contains 5 exons. This protein is expressed in pro-B-cells and is thought to play a role in their differentiation. Using flow cytometric analysis, It has been found that expression of PDCD1 was upregulated on CD16-positive and CD16-negative monocytes, but not on dendritic cells, in viremic HIV-positive patients, but not in highly active antiretroviral therapy (HAART)-treated HIV-positive patients. PDCD1 upregulation in monocytes was induced by microbial Toll-like receptor ligands and inflammatory cytokines. Functional note: Inhibitory cell surface receptor involved in the regulation of T-cell function during immunity and tolerance. Upon ligand binding, inhibits T-cell effector functions in an antigen- specific manner. Possible cell death inducer, in association with other factors. . Reported localization: Membrane; Single-pass type I membrane protein.

Research relevance and current trends

Apoptosis: Researchers commonly examine how PDCD1 (Programmed cell death protein 1) relates to this theme using model systems and orthogonal readouts.

Associated Proteins: Researchers commonly examine how PDCD1 (Programmed cell death protein 1) relates to this theme using model systems and orthogonal readouts.

Developmental Biology: Researchers commonly examine how PDCD1 (Programmed cell death protein 1) relates to this theme using model systems and orthogonal readouts.

Common research applications

IHC/IHC-F: assess spatial distribution of PDCD1 (Programmed cell death protein 1) across tissue regions and cell types using matched controls.

Notes for experimental interpretation

Specificity notes: No cross reactivity with other proteins.

Cross-reactivity: No cross-reactivity with other proteins

Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.

Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

//cdn.shopify.com/s/files/1/0949/7424/7277/files/rp1039-pdcd1-primary-antibodies-ihc-testing-2.jpg?v=1772608088
