Pleiotrophin (PTN), also known as heparin-binding brain mitogen (HBBM) or heparin-binding growth factor 8 (HBGF-8) or neurite growth-promoting factor 1 (NEGF1) or heparin affinity regulatory peptide (HARP) or heparin binding growth associated molecule (HB-GAM), is a protein that in humans is encoded by the PTN gene. PTN is the first member of a family of developmentally regulated cytokines. The PTN gene is mapped to 7q33-q34. A mutant PTN that contained only the first N-terminal 40 amino acids was a dominant negative. Pleiotrophin is expressed in the central and peripheral nervous system and also in several non-neural tissues, notably lung, kidney, gut and bone. Pleiotrophin binds to cell-surface nucleolin as a low affinity receptor. This binding can inhibit HIV infection. Functional note: Secreted growth factor that induces neurite outgrowth and which is mitogenic for fibroblasts, epithelial, and endothelial cells (PubMed:1768439, PubMed:1733956). Binds anaplastic lymphoma kinase (ALK) which induces MAPK pathway activation, an important step in the anti-apoptotic signaling of PTN and regulation of cell proliferation (PubMed:11278720). Binds to cell-surface target proteins via their chondroitin sulfate groups (PubMed:26896299). Down-regulates PTPRZ1 activity (PubMed:16814777). Reported localization: Secreted. Expression/tissue context: Osteoblast and brain.

Research relevance and current trends

Cancer: Researchers commonly examine how PTN relates to this theme using model systems and orthogonal readouts.

Cytokines: Researchers commonly examine how PTN relates to this theme using model systems and orthogonal readouts.

Cytoskeleton/ECM: Researchers commonly examine how PTN relates to this theme using model systems and orthogonal readouts.

Common research applications

Western blotting: compare relative PTN levels across conditions; band patterns may reflect isoforms and processing.

ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

Specificity notes: No cross reactivity with other proteins.

Cross-reactivity: No cross-reactivity with other proteins.

Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.

Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

//cdn.shopify.com/s/files/1/0949/7424/7277/files/A01368-1-Pleiotrophin-primary-antibodies-WB-testing-1.jpg?v=1772608370
