The target protein was coated onto an assay plate overnight at 2 µg\/ml at 4°. The next day, the plate was washed on blocked. Then, serial dilutions of the antibody were added (0 to 200 nM) for 1 hour at room temperature. After washing, HRP-Streptavidin was incubated for 30 minutes at room temperature. Following another wash step, colorimetric HRP substrate was added and quenched with 1N HCL. Then we measured absorbance on a plate reader at 450 nm.
