Sepsis is a morbid condition induced by a toxin, the introduction or accumulation of which is most commonly caused by infection or trauma. Sepsis-inducing toxins have been found associated with pathogenic bacteria, viruses, plants and venoms.Among the well described bacterial toxins are the endotoxins or lipopolysaccharides (LPS) of the gram-negative bacteria. Upon introduction of LPS into the blood it binds to lipopolysaccharide binding protein (LBP). An antibody specific for LBP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyLBP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LBP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LBP bound in the initial step.
