This enzyme is composed of two subunits, a large catalytic subunit (alpha) and a smaller glycoprotein subunit (beta). The beta subunit regulates, through assembly of alpha\/beta heterodimers, the number of sodium pumps transported to the plasma membrane.\u003c\/p\u003e\n\u003ch2\u003eAssay Principle\u003c\/h2\u003e\n\u003cp\u003eThis assay employs a two-site sandwich ELISA to quantitate Na\/K ATPase in samples. An antibody specific for Na\/K ATPase has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNa\/K ATPase present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Na\/K ATPase is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Na\/K ATPase bound in the initial step.
