Originally isolated from porcine aortic endothelial cells, ET-1 is now known to be one of a family of three mammalian vasoactive peptides that also includes Endothelin-2 (ET-2) and Endothelin-3 (ET-3). These related peptides differ from ET-1 at two and six amino acid residue positions, respectively. A fourth peptide, vasoactive intestinal contractor (VIC), is sometimes classified as rat ET-2. All members of the endothelin family contain two essential disulfide bridges and six conserved.\u003c\/p\u003e\n\u003ch2\u003eAssay Principle\u003c\/h2\u003e\n\u003cp\u003eThis assay employs a two-site sandwich ELISA to quantitate EDN1 in samples. An antibody specific for EDN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyEDN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EDN1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EDN1 bound in the initial step.
