It has eight alpha helices and a hydrophobic core. It has a molecular weight of 16,700 daltons, and is the primary oxygen-carrying pigment of muscle tissues. Unlike the blood-borne hemoglobin, to which it is structurally related, this protein does not exhibit cooperative binding of oxygen, since positive cooperativity is a property of multimeric\/oligomeric proteins only. An antibody specific for MB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MB is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MB bound in the initial step.
