This enzyme catalyzes the transient breaking and rejoining of a single strand of DNA which allows the strands to pass through one another, thus reducing the number of supercoils and altering the topology of DNA. This enzyme forms a complex with BLM which functions in the regulation of recombination in somatic cells.High expression is found in testis, heart, skeletal muscle and pancreas. Reduces the number of supercoils in a highly negatively supercoiled DNA.\u003c\/p\u003e\n\u003ch2\u003eAssay Principle\u003c\/h2\u003e\n\u003cp\u003eThis assay employs a two-site sandwich ELISA to quantitate TOP3A in samples. An antibody specific for TOP3A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTOP3A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TOP3A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TOP3A bound in the initial step.
