The pathway activated by binding of Ghrelin to the Growth hormone secretagogue receptor, GHSR1a, regulates the activation of the downstream mitogen-activated protein kinase, Akt, nitric oxide synthase, and AMPK cascad.\u003c\/p\u003e\n\u003ch2\u003eAssay Principle\u003c\/h2\u003e\n\u003cp\u003eThis assay employs a two-site sandwich ELISA to quantitate GHRP-Ghrelin in samples. An antibody specific for GHRP-Ghrelin has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyGHRP-Ghrelin present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GHRP-Ghrelin is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GHRP-Ghrelin bound in the initial step.
