MAP kinases act as an integration point for multiple biochemical signals, and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. This kinase is activated by various environmental stresses and proinflammatory cytokines. The activation requires its phosphorylation by MAP kinase kinases (MKKs), or its autophosphorylation triggered by the interaction of MAP3K7IP1\/TAB1 protein with this kinase. An antibody specific for MAPK14 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAPK14 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAPK14 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAPK14 bound in the initial step.
