MTMR14 is a phosphatidylinositol-3-phosphatase that dephosphorylates the same substrates as myotubularin, PtdIns(4,5)P2 and PtdIns(3,4,5)P3.
MTMR14encodes a myotubularin-related protein. The encoded protein is a phosphoinositide phosphatase that specifically dephosphorylates phosphatidylinositol 3,5-biphosphate and phosphatidylinositol 3-phosphate. Mutations in this gene are correlated with autosomal dominant centronuclear myopathy. Alternate splicing results in multiple transcript variants. A pseudogene of this gene is found on chromosome 18. Expressed in various tissues, including heart, skeletal muscle, placenta, liver, lung, kidney and pancreas.

Assay Principle

This assay employs a two-site sandwich ELISA to quantitate MTMR14 in samples. An antibody specific for MTMR14 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMTMR14 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MTMR14 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MTMR14 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Performance Data

Assay Type

Sandwich ELISA

Detection Range

74.07-6000 pg/mL

Sensitivity

27.38 pg/mL

Species Reactivity

Human (Homo sapiens)

Stability

The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calculated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Manufacturer No.

AE60394HU

UniProt #

Q8NCE2

Alternative Names

C3orf29; FLJ22405; FLJ46453; FLJ90311; HJUMPY; hEDTP; HCV NS5A-transactivated protein 4 splice variant A-binding protein 1|NS5ATP4ABP1|egg-derived tyrosine phosphatase homolog|jumpy

Assay Workflow

The kit includes the components required for standard-curve quantification (see the kit manual for the component table and validated sample types).
