Granins may serve as precursors for biologically active peptides. Some granins have been shown to function as helper proteins in sorting and proteolytic processing of prohormones; however, the function of this protein is unknown. The deduced 468-amino acid protein, which has a calculated molecular mass of 53 kD, contains an N-terminal signal peptide, a secretogranin motif, 3 potential N-glycosylation sites, and a putative tyrosine sulfation site. SCG3 shares 87%, 87%, and 56% overall id.\u003c\/p\u003e\n\u003ch2\u003eAssay Principle\u003c\/h2\u003e\n\u003cp\u003eThis assay employs a two-site sandwich ELISA to quantitate SCG3 in samples. An antibody specific for SCG3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySCG3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SCG3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SCG3 bound in the initial step.
