The protein localizes to the cytoplasm and the plasma membrane of premeiotic testicular germ cells and it may be involved in progression of testicular tumors of germ cell origin. The deduced 314-amino acid ESP1 precursor protein has a calculated molecular mass of 33.1 kD, and the 273-amino acid active form has a calculated molecular mass of 30.6 kD. An antibody specific for PRSS21 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPRSS21 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PRSS21 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRSS21 bound in the initial step.
