It belongs to the TMTC family and contains 10 TPR repeats. The tetratrico peptide repeat (TPR) is a structural motif present in a wide range of proteins. It mediates protein-protein interactions and the assembly of multiprotein complexes. The TPR motif consists of 3-16 tandem-repeats of 34 amino acids residues, although individual TPR motifs can be dispersed in the protein sequence. Sequence alignment of the TPR domains reveals a consensus sequence defined by a pattern of small and large amino acids. An antibody specific for TMTC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTMTC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TMTC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMTC1 bound in the initial step.
