RNA dot blot analysis detected expression in all tissues examined. Fluorescence-tagged TMEM55B localized with membrane markers of late endosomes or lysosomes. Endogenous HeLa cell TMEM55B showed a similar distribution.It showed no activity toward other phosphatidylinositol substrates or inositol phosphates tested. After removing any unbound substances, a biotin-conjugated antibody specific for TMEM55B is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMEM55B bound in the initial step.
