Tryptases are enzymatically active only as heparin-stabilized tetramers, and they are resistant to all known endogenous proteinase inhibitors. The alleles of this gene exhibit an unusual amount of sequence variation, such that the alleles were once thought to represent two separate genes, beta II and beta III. An antibody specific for TPSB2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTPSB2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TPSB2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TPSB2 bound in the initial step.
