It is located in the rim regions of the flattened disks that contain rhodopsin, which is the protein that is responsible for initiation of visual phototransduction upon reception of light. Peripherin 2 may function as an adhesion molecule involved in stabilization and compaction of outer segment disks or in the maintenance of the curvature of the rim. This protein is essential for disk morphogenesis. Defects in this gene are associated with both central and peripheral retinal degene.\u003c\/p\u003e\n\u003ch2\u003eAssay Principle\u003c\/h2\u003e\n\u003cp\u003eThis assay employs a two-site sandwich ELISA to quantitate PRPH2 in samples. An antibody specific for PRPH2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPRPH2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PRPH2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRPH2 bound in the initial step.
