It is highly expressed in the outer hair cells, and is not expressed in the nonmotile inner hair cells. Immunolocalization shows prestin is expressed in the lateral plasma membrane of the outer hair cells, the region where electromotility occurs. The expression pattern correlates with the appearance of outer hair cell electromotility. Prestin is specifically expressed in outer hair cells (OHCs) of the cochlea and is essential in auditory processing. An antibody specific for SLC26A5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySLC26A5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SLC26A5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SLC26A5 bound in the initial step.
