Methylation patterns are responsible for the repression of parasitic sequence elements and the expression status of genes subject to genomic imprinting and X inactivation. Faithful maintenance of methylation patterns is required for normal mammalian development, and aberrant methylation patterns are associated.\u003c\/p\u003e\n\u003ch2\u003eAssay Principle\u003c\/h2\u003e\n\u003cp\u003eThis assay employs a two-site sandwich ELISA to quantitate DNMT1 in samples. An antibody specific for DNMT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDNMT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DNMT1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DNMT1 bound in the initial step.
