It was first described in the cytosol of rabbit brain as an oligopeptidase, which degrades the nonapeptide bradykinin at the Pro-Phe bond.[5] The enzyme is involved in the maturation and degradation of peptide hormones and neuropeptides such as alpha-melanocyte-stimulating hormone, luteinizing hormone-releasing hormone (LH-RH), thyrotropin-rele.\u003c\/p\u003e\n\u003ch2\u003eAssay Principle\u003c\/h2\u003e\n\u003cp\u003eThis assay employs a two-site sandwich ELISA to quantitate PREP in samples. An antibody specific for PREP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPREP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PREP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PREP bound in the initial step.
