The plate has been coated with sheep polyclonal antibody against mouse NGF. Any NGF present in the sample or standard will be captured. A biotin-conjugated anti-murine NGF antibody is added as a detection antibody along with streptavidin-horseradish peroxidase (HRP) and an accompanying substrate. The reaction exhibits a blue color in direct proportion to the amount of NGF in the initial sample. The reaction is terminated by the addition of a stop solution and absorbance is measured at 450nm.
