Product Overview
Clone ABFB-16 is a rabbit monoclonal antibody that recognizes proprotein convertase subtilisin/kexin type 9 (gene PCSK9) in human samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 74.3 kDa, and the supplier reports an observed band at about 75 kDa.
PCSK9 is a secreted proprotein convertase made mainly in the liver that binds the LDL receptor and sends it for lysosomal degradation. By lowering LDL receptor levels it raises plasma LDL cholesterol, which made PCSK9 a major target of cholesterol-lowering antibodies and siRNA drugs.

Samples with a confirmed band (WB): human A431 cells, human HeLa cells, human HepG2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Reactivity Notes
The supplier lists reactivity with human. UniProt describes proprotein convertase subtilisin/kexin type 9 as secreted and also located in the cytoplasm, endosome and lysosome. Tissue expression noted by UniProt (human): Expressed in neuro-epithelioma, colon carcinoma, hepatic and pancreatic cell lines, and in Schwann cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
