Chloroquine diphosphate is a lysosomotropic agent widely used to inhibit autophagy by preventing lysosomal acidification and autophagosome-lysosome fusion. In neuroscience, chloroquine is employed to study autophagic flux and lysosomal function in neurodegenerative disease models. Impaired autophagy is a hallmark of disorders such as Alzheimer’s and Parkinson’s, where the accumulation of protein aggregates and damaged organelles contributes to neuronal dysfunction. Chloroquine’s ability to modulate autophagy and endolysosomal pathways provides insights into the cellular mechanisms underlying neurodegeneration. Additionally, its anti-inflammatory properties have been explored in the context of neuroinflammation and glial cell regulation.
