Puromycin is a broad-spectrum antibiotic and potent inhibitor of protein synthesis. It mimics the 3' end of aminoacyl-tRNA and incorporates into the ribosome's A site, causing premature termination of translation. This mechanism disrupts polypeptide elongation, making puromycin a valuable tool for studying protein synthesis and turnover in both prokaryotic and eukaryotic systems. In neuroscience, puromycin is frequently used to investigate synaptic plasticity, neuronal translation dynamics, and the regulation of local protein synthesis in dendrites and axons—processes critical to memory formation and neurodevelopment.
