The catch is specificity. Retatrutide, tirzepatide, semaglutide, dulaglutide, and liraglutide are all fatty acid–modified peptides built on a related incretin backbone. An antibody raised against one may recognize others, and it may also recognize endogenous GLP-1 or GIP in your matrix. If your study design involves a comparator arm, a switch, or an animal model with meaningful endogenous incretin tone, an assay you have not characterized will return numbers that look clean and mean something other than what you think.

Retatrutide Half-Life, Albumin Binding, and What They Mean for Your Assay
Retatrutide (LY3437943) is a single fatty diacid–conjugated peptide with agonist activity at the GIP, GLP-1, and glucagon receptors. In vitro it shows relatively balanced glucagon and GLP-1 receptor activity with greater potency at the GIP receptor (Coskun et al., Cell Metab 2022). The retatrutide half-life is approximately six days with dose-proportional pharmacokinetics, which supports once-weekly subcutaneous dosing (Urva et al., Lancet 2022).
Three features make lipidated incretin peptides demanding to measure:
- Drug-specific detection is required. Retatrutide shares design features with other incretin-based agents, so a generic GLP-1 immunoassay will not give you drug concentration. PK kits solve this with antibody pairs raised against the specific drug, which is why each analog has its own dedicated kit rather than a shared one.
- Lipidation and protein binding. The fatty diacid moiety drives albumin binding, which extends half-life but also means a fraction of your analyte is complexed in plasma. Sample handling and dilution buffer can shift the free/bound equilibrium and therefore the apparent concentration.
- Long half-life, wide dynamic range. A six-day half-life over a multi-week study means samples span a large concentration range. A single dilution factor will not keep every timepoint on the linear part of the curve.

How to Qualify a Retatrutide ELISA Kit: Five Checks Before You Report

1. Confirm the standard curve range fits your study, not the other way around

2. Run dilution linearity in your actual matrix
Serial-dilute a high-concentration study sample (not a spiked buffer standard) across at least four dilutions and back-calculate. Non-parallelism between your sample curve and the standard curve is the single most informative red flag in peptide bioanalysis — it usually indicates matrix interference or a binding-protein effect, and it invalidates the assumption that one dilution factor applies to all timepoints.

3. Establish spike recovery per species and per matrix
The kit specifies typical recovery of 80–120%. Confirm that in your own species and anticoagulant. Mouse plasma, rat serum, and NHP plasma are not interchangeable matrices, and EDTA, heparin, and citrate can each behave differently with a lipidated peptide. Spike at low, mid, and high points on the curve.
4. Qualify selectivity for multi-analog study designs
Drug-specific kits are built to discriminate their analyte, and for a single-agent study the analyte-matched kit is the answer. If your design puts two analogs in the same program — a comparator arm, a crossover, or a switch study — running each related peptide through each assay once at pharmacologically relevant concentrations documents selectivity for your specific pairing and takes an afternoon. It is standard method qualification, and reviewers expect to see it.
5. Bridge to an orthogonal method for anything that leaves your lab
For regulated bioanalysis, or for any dataset supporting a dose-selection or comparability claim, cross-validate a subset of samples by LC-MS/MS. ELISA is well suited to screening, exploratory PK, and relative comparisons across arms; it is not a substitute for a validated mass-spec method when the numbers carry regulatory weight.

Practical note. Freeze–thaw stability is the most commonly skipped experiment in peptide PK and the most commonly cited weakness afterward. Characterize at least three cycles in your own matrix before you commit a study's worth of samples to a single storage plan.

Catalog gaps — stated up front. Two things researchers ask for that we do not currently stock:
Anti-drug antibody (ADA) assays. The kits above measure drug concentration, not immunogenicity. A bridging ADA assay is a separate reagent set and is not currently in the BioHippo catalog for these analytes. If your protocol includes an immunogenicity endpoint, plan for a second sourcing path.
Several incretin analogs have no matched PK kit. Exenatide and pramlintide are available as recombinant peptides for use as standards or in functional assays, but not as PK ELISA kits. Cagrilintide, survodutide, and mazdutide are not currently represented in either form.
FAQ: Peptide Drug Quantification by ELISA
Can I use a retatrutide ELISA kit to measure tirzepatide or semaglutide?
Use the analyte-matched kit. These are drug-specific assays — each analog has its own antibody pair and its own SKU precisely because the kits are built to detect one drug rather than the class. A retatrutide kit is designed to measure retatrutide, and BioHippo stocks dedicated kits for tirzepatide, semaglutide, dulaglutide, liraglutide, and orforglipron alongside it. If a single study spans more than one analog, document selectivity for that specific pairing as part of your method qualification.
ELISA or LC-MS/MS for preclinical incretin PK?
ELISA is the practical choice for exploratory PK, dose-ranging, relative comparisons between arms, and any situation where throughput matters more than absolute accuracy. LC-MS/MS is the right call when you need unambiguous specificity against structurally related compounds, when the data supports a regulatory submission, or when you are working below the immunoassay's LLOQ. Many programs use both: ELISA for the bulk of samples, mass spec to cross-validate a subset.
Does albumin binding interfere with the measurement?
It can. Fatty acid–modified peptides bind albumin extensively, and an immunoassay measures whatever fraction the antibody can access under the assay's conditions. Dilution into assay buffer shifts that equilibrium, which is one reason dilution linearity in your own matrix is more informative than a spiked-buffer recovery experiment. Report your minimum required dilution alongside your concentrations so results are interpretable.
Will endogenous GLP-1 or GIP show up in my baseline samples?
It is unlikely to be a practical problem. Endogenous incretins circulate in the low pg/mL range while these assays operate in ng/mL, roughly three orders of magnitude apart, and the antibody pairs are raised against the drug rather than the native hormone. Including predose and vehicle-control wells is still worth doing — it establishes your true assay background for that species and matrix, which you will want when interpreting terminal timepoints near the LLOQ.
Can these kits be used on human clinical samples?
These are research use only products. They are not validated or intended for diagnostic use, clinical decision-making, therapeutic drug monitoring in patients, or verification of material obtained outside a licensed supply chain. Use in a clinical study requires the appropriate regulatory framework and, in most cases, a validated method rather than an RUO kit.
How many plates should I budget for a rodent PK study?
Work backwards from your sampling schedule. A 96-well plate accommodates roughly 36–40 samples in duplicate once you allow for a standard curve and QC wells. With a six-day half-life you will want dense early sampling plus a long terminal tail, so multiply your animal count by timepoints and add 20–30% for reruns and dilution repeats. Order enough kit lots at once to avoid a lot change mid-study, or plan a bridging experiment if you cannot.
