Overview
Normal Human IgG Isotype Control Antibody, FITC is designed to match primary antibody format for background estimation. Commonly used in FCM, Isotype Control applications. Typical workflows include Select staining controls in cells/tissues (isotype/KD/KO), Phenotype target-positive cells by flow cytometry in PBMCs/cell suspensions, Optimize antibody dilution for staining in cells/tissues using a reference sample, QC lot-to-lot by comparing signal in a reference lysate or cell line. Isotype control rationale
Isotype control antibodies are matched to the primary antibody’s species and isotype, but lack specificity for a cellular antigen. They are used to estimate non‑specific binding and background signal in immunoassays. In flow cytometry and immunofluorescence, isotype controls can help identify staining driven by Fc receptor interactions, sticky epitopes, or fluorophore/label-associated background rather than true antigen recognition. When conjugated, the isotype control should be used with the same fluorophore/label and at the same concentration as the corresponding test antibody. This reagent is characterized by its host species (Human), isotype (IgG), which should match your primary antibody format as closely as possible.
How this reagent helps
Use this isotype control to estimate non-specific background under matched staining conditions. For best performance, interpret signal in the context of appropriate controls and orthogonal validation when available.
Recommended experimental notes
- Titrate the antibody/reagent to optimize signal-to-noise for your specific sample type and detection system.
- Run the isotype control at the same concentration, incubation time, and staining conditions as the test antibody.
- For flow cytometry, consider Fc blocking and use an FMO (fluorescence minus one) control to support gating decisions.
- Include appropriate controls (e.g., unstained, secondary-only where relevant, and biological positives/negatives).
- For flow cytometry, include Fc blocking when applicable, use a viability dye, and consider staining live cells for surface markers prior to fixation/permeabilization.
