2× Hieff™ Ultra-Rapid II HotStart Universal PCR Master Mix contains antibody-modified heat-stabilized Taq DNA Polymerase with a strong elongation factor and optimized buffer system for ultra-high amplification efficiency. Particularly suited for direct colony PCR from E. coli, Agrobacterium, and yeast. Pre-loaded red tracer dye enables direct gel electrophoresis. Validated shelf life of 2 years at −20°C.

This ultra-rapid mix achieves 1 sec/kb for simple templates such as plasmids up to 6 kb. For complex templates, extension rates are 1 sec/kb for ≤3 kb, 3 sec/kb for 3–6 kb, 5 sec/kb for 6–10 kb, and 10 sec/kb for >10 kb fragments from genomic DNA.

The heat-stabilized Taq DNA Polymerase is modified with monoclonal antibodies that block polymerase activity at ambient temperature. At initial denaturation (≥95°C), the antibodies dissociate irreversibly, releasing fully active enzyme. This prevents non-specific priming during reaction assembly.

Yes. This mix is specifically validated for direct colony PCR from E. coli, Agrobacterium, and yeast. Colony material can be added directly to the reaction without prior culture preparation or DNA extraction.

No. The mix contains a pre-loaded red tracer dye enabling direct gel loading of PCR products immediately after amplification.

The formulation contains a protective agent validated for 2 years of stability at −25 to −15°C. High stability is confirmed by the manufacturer for the full stated shelf life.
