Enzyme & Substrate
S-adenosylhomocysteine hydrolase is the only mammalian enzyme that clears SAH, and because SAH inhibits virtually every methyltransferase, its activity sets the cell's overall methylation capacity.
Assay Principle
The Adenosyl Homocysteinase (AHCY) Fluorometric Assay Kit applies a fluorometric format to this measurement, read at Ex/Em = 400/465 nm. Hands-on and incubation time is 60 minutes.
Adenosylhomocysteinase (AHCY) or S-adenosylhomocysteine hydrolase (SAHH) is an enzyme that catalyzes the reversible hydrolysis of S-Adenosyl Homocysteine (SAH) to adenosine and homocysteine (Hcy).
Measuring AHCY activity reports the cell's capacity to clear SAH and therefore its methylation headroom.
