Biological Activity

Alaproclate is a selective serotonin reuptake inhibitor (SSRI).1,2 It inhibits depletion of serotonin (5-HT) induced by 4-methyl-alpha-ethyl-m-tyramine in rat cerebral cortex, hippocampus, hypothalamus, and striatum (EC50s = 18, 4, 8, and 12 mg/kg, respectively).1 Alaproclate inhibits NMDA-evoked currents and depolarization-induced voltage-dependent potassium currents in rat hippocampal neurons (IC50s = 1.1 and 6.9 uM, respectively) and does not inhibit GABA-evoked currents when used at concentrations up to 100 uM.2 It increases sirtuin 1 (SIRT1) levels in N2a murine neuroblastoma cells expressing apolipoprotein E4 (ApoE4; IC50 = 2.3 uM) and in the hippocampus in the FXFAD-ApoE4 transgenic mouse model of Alzheimer's disease when administered at a dose of 20 mg/kg twice daily.3 Alaproclate (40 mg/kg) decreases immobility time in the forced swim test in rats, indicating antidepressant-like activity.4

What should I do if the products in powder form adhere to the vial tightly?

Centrifuge the closed vial at 3000 rpm for 1–2 minutes to consolidate the powder at the bottom. For electrostatically charged powders, briefly warm the vial to room temperature and allow it to equilibrate before opening. Then add the appropriate solvent directly onto the powder and vortex before sonicating. Avoid tapping the vial vigorously, as this can generate fine powder aerosol.

After the inhibitor is dissolved in DMSO to prepare the stock solution, can it be stored at room temperature for one week?

It is not recommended to store DMSO stock solutions at room temperature. Once the stock solution is prepared, aliquot into single-use volumes and store at −20 °C (or −80 °C for sensitive compounds). Room-temperature storage accelerates chemical degradation and moisture uptake. When ready to use, thaw an aliquot at room temperature, use immediately, and avoid repeated freeze-thaw cycles.
