  • Molecular weight guidance: Observed: 142 kDa; Calculated: 141523 MW

Specificity note: No cross reactivity with other proteins.

Biological background

Protein function (datasheet): Mediates ATP-dependent export of organic anions and drugs from the cytoplasm. Hydrolyzes ATP with low efficiency. Not capable of conferring drug resistance. Mediates the translocation of phosphatidylcholine across the canalicular membrane of the hepatocyte.

Scientific background (datasheet): Adenosine triphosphate-binding cassette subfamily B, member 4 (ABCB4), also called MDR3, is a protein that in humans is encoded by the ABCB4 gene. MDR1 and MDR3 constitute the human P-glycoprotein gene family. The MDR3 gene contains 28 exons and 27 of these contain coding sequences for the two homologous halves of the protein that correlate with functional domains. MDR3 gene mutations represent a genetic factor involved in this peculiar form of cholesterol gallstone disease in adults.

Cellular localization (datasheet): Cell membrane; Multi-pass membrane protein.

Tissue details (datasheet): Expressed in heart, kidney, lung, pancreas, placenta, ovary, small intestine and spleen. Isoform 2 is expressed predominantly in scirrhous gastric carcinoma and, weakly in placenta. Overexpression is associated with several cancers including breast cancer and colon tumors. Isoform 2 is overexpressed in scirrhous gastric carcinoma.

Sequence similarities (datasheet): Belongs to the ABC transporter superfamily. ABCB family. Multidrug resistance exporter (TC 3.A.1.201) subfamily.

Research relevance and current trends

  • Commonly studied in contexts related to ATPases,Cancer,Lipid Metabolism,Metabolism,Nuclear Import/Export,Plasma Membrane,Protein Trafficking,Signal Transduction.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
