Overview

This antibody is intended for detection of ACTA1 (Actin, alpha skeletal muscle) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-Actin/ACTA1 Antibody catalog # RP1070. Tested in IHC, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human Actin, identical to the related mouse and rat sequences.
  • Molecular weight context: reported MW: 42 kDa; calculated MW: 42051 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Actin, alpha skeletal muscle; Actin, alpha skeletal muscle. Actin, a highly conserved protein, is a major component of both the cytoskeletal and contractile structures in the cell types. It varies in amount, being related to the type of differentiation and to the functional state of cells and tissues. The actins exhibit over 90% sequence homology, but each isoform has a unique NH2-terminal sequence. The isoforms are comprised of three alpha-actin, one beta-actin, two gamma-actin. Because the amino acid sequence of the C-terminal is the same for almost all actins, this antibody has been raised using a synthetic peptide corresponding to the C-terminal 11 residues. Functional note: Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells. Reported localization: Cytoplasm, cytoskeleton. Expression/tissue context: Expressed in cerebral cortex. .

Research relevance and current trends

  • Actin: Researchers commonly examine how ACTA1 (Actin, alpha skeletal muscle) relates to this theme using model systems and orthogonal readouts.
  • Actin Assembly: Researchers commonly examine how ACTA1 (Actin, alpha skeletal muscle) relates to this theme using model systems and orthogonal readouts.
  • Actin Binding Proteins: Researchers commonly examine how ACTA1 (Actin, alpha skeletal muscle) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative ACTA1 (Actin, alpha skeletal muscle) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of ACTA1 (Actin, alpha skeletal muscle) across tissue regions and cell types using matched controls.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Family / similarity context: Belongs to the synaptopodin family.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

We were happy with the WB result of your anti-Actin/ACTA1 antibody. However we have been able to see positive staining in biceps brachii cytoskeleton. using this antibody. Is that expected? Could you tell me where is ACTA1 supposed to be expressed?
According to literature, biceps brachii does express ACTA1. Generally ACTA1 expresses in cytoplasm, cytoskeleton. Regarding which tissues have ACTA1 expression, here are a few articles citing expression in various tissues: Skeletal muscle, Pubmed ID: 6190133, 15489334
Does RP1070 anti-Actin/ACTA1 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
You can see on the product datasheet, RP1070 anti-Actin/ACTA1 antibody as been validated on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
