Biological background

Protein function (datasheet): Converts angiotensin I to angiotensin II by release of the terminal His-Leu, this results in an increase of the vasoconstrictor activity of angiotensin. Also able to inactivate bradykinin, a potent vasodilator. Has also a glycosidase activity which releases GPI-anchored proteins from the membrane by cleaving the mannose linkage in the GPI moiety.

Scientific background (datasheet): Angiotensin I converting enzyme (ACE), also called DCP or CD143, is a zinc-containing dipeptidyl carboxypeptidase widely distributed in mammalian tissues and is though to play a critical role in blood pressure regulation. ACE gene is mapped to 17q23.3. This gene encodes an enzyme involved in catalyzing the conversion of angiotensin I into a physiologically active peptide angiotensin II. Angiotensin II is a potent vasopressor and aldosterone-stimulating peptide that controls blood pressure and fluid-electrolyte balance. This enzyme plays a key role in the renin-angiotensin system. Many studies have associated the presence or absence of a 287 bp Alu repeat element in this gene with the levels of circulating enzyme or cardiovascular pathophysiologies.

Cellular localization (datasheet): Angiotensin-converting enzyme, soluble form: Secreted.

Tissue details (datasheet): Ubiquitously expressed, with highest levels in lung, kidney, heart, gastrointestinal system and prostate. Isoform Testis-specific is expressed in spermatocytes and adult testis. .

Sequence similarities (datasheet): Belongs to the peptidase M2 family.

Research relevance and current trends

  • Commonly studied in contexts related to Cancer,Cancer Metabolism,Cardiovascular,Cell Biology,Cofactors, Vitamins/Minerals,Hematopoietic Progenitors,Metabolism,Metabolism Processes,Metalloprotease,Pathways and Processes,Proteolysis/Ubiquitin,Proteolytic Enzymes,Response To Hypoxia,Serum Proteins,Signal Transduction,Stem Cells,Surface Molecules,Vasculature,Vitamins/Minerals.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.

We have seen staining in human lung. What should we do? Is anti-Angiotensin Converting Enzyme 1/ACE antibody supposed to stain lung positively?
From what I have seen in literature lung does express ACE. From what I have seen in Uniprot.org, ACE is expressed in testis, brain, lung, umbilical vein endothelial cell, plasma, liver, among other tissues. Regarding which tissues have ACE expression, here are a few articles citing expression in various tissues: Brain, Pubmed ID: 16625196 Liver, Pubmed ID: 19159218 Lung, Pubmed ID: 2558109 Plasma, Pubmed ID: 16335952 Testis, Pubmed ID: 14702039 Umbilical vein endothelial cell, Pubmed ID: 9642152
My question regards using your anti-Angiotensin Converting Enzyme 1/ACE antibody for hormone catabolic process studies. Has this antibody been tested with western blotting on hela whole cell lysate? We would like to see some validation images before ordering.
I appreciate your inquiry. This PA2196-1 anti-Angiotensin Converting Enzyme 1/ACE antibody is tested on human placenta tissue, prostatic cancer tissue, hela whole cell lysate, a549 whole cell lysate, 22rv1 whole cell lysate. It is guaranteed to work for IHC, WB in human. Our Boster guarantee will cover your intended experiment even if the sample type has not been be directly tested.
