Overview

This antibody is intended for detection of ANXA1 (Annexin A1) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-Annexin A1/ANXA1 Antibody Picoband® catalog # PB9127. Tested in IHC, WB applications. This antibody reacts with Human, Monkey, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E.coli-derived human Annexin A1 recombinant protein (Position: A2-N346). Human Annexin A1 shares 88% and 89% amino acid (aa) sequences identity with mouse and rat Annexin A1, respectively. (reported region: A2-N346).
  • Molecular weight context: reported MW: 39 kDa; calculated MW: 38714 MW
  • Reactivity: Human,Monkey,Rat
  • Applications: IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Annexin A1; Annexin A1. ANXA1, also known as lipocortin I or Annexin A1, is a protein that in humans is encoded by the ANXA1 gene. It is mapped to 9q21.13. ANXA1 belongs to a family of Ca (2+)-dependent phospholipid binding proteins which have a molecular weight of approximately 35,000 to 40,000 and are preferentially located on the cytosolic face of the plasma membrane. ANXA1 protein has an apparent relative molecular mass of 40 kDa, with phospholipase A2 inhibitory activity. Lower peptide concentrations possibly found in inflammatory situations elicit Ca (2+) transients without fully activating the mitogen-activated protein kinase pathway. This causes a specific inhibition of the transendothelial migration of neutrophils and a desensitization of neutrophils toward a chemoattractant challenge. These findings identified ANXA1 peptides as novel, endogenous FPR ligands and established a mechanistic basis of ANXA1-mediated antiinflammatory effects. Functional note: Calcium/phospholipid-binding protein which promotes membrane fusion and is involved in exocytosis. This protein regulates phospholipase A2 activity. It seems to bind from two to four calcium ions with high affinity. Reported localization: Nucleus . Cytoplasm . Cell projection, cilium . Basolateral cell membrane . Found in the cilium, nucleus and basolateral cell membrane of ciliated cells in the tracheal endothelium. Found in the cytoplasm of type II pneumocytes and alveolar macrophages. . Expression/tissue context: Ubiquitously expressed, with highest levels in lung, kidney, heart, gastrointestinal system and prostate. Isoform Testis-specific is expressed in spermatocytes and adult testis. .

Research relevance and current trends

  • Calcium Signaling: Researchers commonly examine how ANXA1 (Annexin A1) relates to this theme using model systems and orthogonal readouts.
  • Cell Adhesion: Researchers commonly examine how ANXA1 (Annexin A1) relates to this theme using model systems and orthogonal readouts.
  • Cytoskeleton/ECM: Researchers commonly examine how ANXA1 (Annexin A1) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative ANXA1 (Annexin A1) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of ANXA1 (Annexin A1) across tissue regions and cell types using matched controls.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Family / similarity context: Belongs to the annexin family.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
