Biological background
Protein function (datasheet): This protein is an anticoagulant protein that acts as an in inhibitor of the thromboplastin-specific complex, which is involved in the blood coagulation cascade.
Scientific background (datasheet): Annexin V also know as endonnexin II (ENX2),or placental protein 4 (PP4). Endonexin II is a member of the family of Ca (2+)-dependent phospholipid binding proteins, known as annexins. It bind to the phospholipids that are preferentially located on the cytosolic face of the plasma membrane. It has a relative molecular weight of about 35,000. The gene lies on mouse chromosome 3 in close linkage with the fibroblast growth factor 2 (basic) gene and is syntenic with other genes known to have orthologous counterparts on human chromosome 4q. The PP4 cDNA encoded a protein of 320 amino acid residues. A single mRNA, approximately 1.6 kb long, was found to be expressed in human cell lines and placenta. PP4 is an anticoagulant protein that acts as an in inhibitor of the thromboplastin-specific complex, which is involved in the blood coagulation cascade.
Sequence similarities (datasheet): Belongs to the annexin family.
Research relevance and current trends
- Commonly studied in contexts related to Cell Biology,Cell Death,Invasion/Microenvironment.
- Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
- Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).
Common research applications
- Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
- ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
- Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
- Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
- Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.
Notes for experimental interpretation
- Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
- Cross-reactivity (datasheet): No cross-reactivity with other proteins
- Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.
As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
