Biological background
Protein function (datasheet): This protein is an anticoagulant protein that acts as an in inhibitor of the thromboplastin-specific complex, which is involved in the blood coagulation cascade.
Scientific background (datasheet): Annexin A5 (or annexin V) is a cellular protein in the annexin group. It is mapped to 4q27. Annexin A5 has been proposed to play a role in the inhibition of blood coagulation by competing for phosphatidylserine binding sites with prothrombin and also to inhibit the activity of phospholipase A1. This protein is an anticoagulant protein, it can form a shield around negatively-charged phospholipid molecules, the formation of an annexin A5 shield blocks the entry of phospholipids into coagulation (clotting) reactions. And Annexin A5 can also act as an in inhibitor of the thromboplastin-specific complex, which is involved in the blood coagulation cascade.
Cellular localization (datasheet): Cytoplasm. Nucleus. Cell membrane; Peripheral membrane protein. Localizes within both nucleus and cytoplasm of proliferative primary myoblasts and mostly within the nucleus of differentiated primary myoblasts. By virtue of the N-terminal PH domain, is recruited to sites of the plasma membrane containing increased PI (3,4,5)P3 or PI (3,4)P2, cell membrane targeting is also facilitared by interaction with CLIP3.
Tissue details (datasheet): Expressed in all cell types so far analyzed.
Sequence similarities (datasheet): Belongs to the annexin family.
Research relevance and current trends
- Commonly studied in contexts related to Apoptosis,Apoptotic Markers,Atherosclerosis,Cancer,Cardiovascular,Cell Biology,Cell Death,Invasion/Microenvironment.
- Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
- Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).
Common research applications
- Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
- ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
- Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
- Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
- Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.
Notes for experimental interpretation
- Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
- Cross-reactivity (datasheet): No cross-reactivity with other proteins
- Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.
As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
