Biological background
PNPLA2 (protein: Lysosome-associated membrane glycoprotein 2 (Lamp2)) is a commonly studied target in molecular and cellular biology. Functional context (as provided): One of the major pre-mRNA-binding proteins. Binds tenaciously to poly (C) sequences. Likely to play a role in the nuclear metabolism of hnRNAs, particularly for pre-mRNAs that contain cytidine-rich sequences. Can also bind poly (C) single- stranded DNA. Plays an important role in p53/TP53 response to DNA damage, acting at the level of both transcription activation and repression. When sumoylated, acts as a transcriptional coactivator of p53/TP53, playing a role in p21/CDKN1A and 14-3-3 sigma/SFN induction (By similarity). As far as transcription repression is concerned, acts by interacting with long intergenic RNA p21 (lincRNA-p21), a non-coding RNA induced by p53/TP53. This interaction is necessary for the induction of apoptosis, but not cell cycle arrest. . Reported cellular localization context: Cytoplasm . Nucleus, nucleoplasm . Cell projection, podosome . Recruited to p53/TP53- responsive promoters, in the presence of functional p53/TP53 (PubMed:16360036). In case of ASFV infection, there is a shift in the localization which becomes predominantly nuclear (PubMed:18775702). Tissue expression notes (as provided): Expressed in brain and lymphoblast.
Research relevance and current trends
- Research context keywords from the source record include: Chromatin Binding Proteins,DNA/RNA,DNA/RNA Binding,Epigenetics and Nuclear Signaling,RNA Processing,Splicing.
- Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
- Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.
Common research applications
- Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
Workflow ideas (metafield): Validate PNPLA2 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect PNPLA2 expression by Western blot in cell or tissue lysates, Compare relative PNPLA2 levels across experimental conditions (dose/time-course) using antibody-based readouts
Notes for experimental interpretation
- Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
- Apparent molecular weight may vary by sample type and processing (observed MW: 55 kDa; calculated MW: 50976 MW).
- Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.
