Product Overview
This rabbit polyclonal antibody detects catenin beta-1 (gene CTNNB1) in rat, mouse and human samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 85.5 kDa, and the supplier reports an observed band at about 95 kDa.
β-Catenin (CTNNB1) has two roles: it is a structural part of cadherin-based adherens junctions and the central effector of canonical Wnt signaling. Without Wnt, a destruction complex of APC, AXIN, CK1α and GSK3B targets it for proteasomal degradation; with Wnt, it builds up in the nucleus and activates TCF/LEF target genes. It is studied in development, stem cells and cancer.

|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
| IHC (frozen sections) | 0.5–1 µg/mL (Mouse, Rat, Human) |

Samples with a confirmed band (WB): MM453 cells, MCF-7 cells, HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Samples with confirmed staining (IHC): human mammary cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (IHC): rat intestine tissue.
Samples with confirmed staining (ICC/IF): A431 cells.
Immunogen
Synthetic peptide from the C-terminal region of human beta-catenin. The peptide sequence is conserved in rat and mouse.
Reactivity Notes
The supplier lists reactivity with rat, mouse and human. UniProt places catenin beta-1 in the cytoplasm, nucleus, cell junction and cell membrane. Tissue expression noted by UniProt (human): Expressed in several hair follicle cell types: basal and peripheral matrix cells, and cells of the outer and inner root sheaths. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
