Vendor description (summary): Boster Bio Anti-beta glucuronidase/GUSB Antibody Picoband® catalog # A02234-1.
Biological background
Biological context: Plays an important role in the degradation of dermatan and keratan sulfates.
Expression and localization notes: cellular localization: Membrane. Single-pass type I membrane protein., tissue context: Broadly expressed with higher expression in heart..
Common research applications
- Western blotting (WB): Compare GUSB levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of GUSB in tissue sections, considering fixation and antigen retrieval effects.
- Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
- Flow cytometry: Quantify GUSB-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Specificity: No cross reactivity with other proteins.
- Background: Beta-glucuronidases are members of the glycosidase family of enzymes that catalyze breakdown of complex carbohydrates. This gene encodes a hydrolase that degrades glycosaminoglycans, including heparan sulfate, dermatan sulfate, and chondroitin-4,6-sulfate. The enzyme forms a homotetramer that is localized to the lysosome. Mutations in this gene result in mucopolysaccharidosis type VII. Alternative splicing results in multiple transcript variants. There are many pseudogenes of this locus in the human genome.
- Cross reactivity: No cross-reactivity with other proteins.
- Cellular localization: Membrane. Single-pass type I membrane protein.
- Tissue details: Broadly expressed with higher expression in heart.
