Vendor description (summary): Boster Bio Anti-BHC80/PHF21A Antibody catalog # A08507-1.

Biological background

Biological context: Component of the BHC complex, a corepressor complex that represses transcription of neuron-specific genes in non-neuronal cells. The BHC complex is recruited at RE1/NRSE sites by REST and acts by deacetylating and demethylating specific sites on histones, thereby acting as a chromatin modifier. In the BHC complex, it may act as a scaffold. Inhibits KDM1A-mediated demethylation of 'Lys-4' of histone H3 in vitro, suggesting a role in demethylation regulation.

Expression and localization notes: cellular localization: Nucleus., tissue context: Highly expressed in brain. Expressed at much lower level in other tissues..

Common research applications

  • Western blotting (WB): Compare PHF21A levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of PHF21A in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify PHF21A-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: PHD finger protein 21A is a protein that in humans is encoded by the PHF21A gene. The PHF21A gene encodes BHC80, a component of a BRAF35 (MIM 605535)/histone deacetylase (HDAC; see MIM 601241) complex (BHC) that mediates repression of neuron-specific genes through the cis-regulatory element known as repressor element-1 (RE1) or neural restrictive silencer (NRS).
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Nucleus.
  • Tissue details: Highly expressed in brain. Expressed at much lower level in other tissues.
  • Research category: Cancer,Cell Biology,Cytoskeleton/ECM,ECM Enzymes,Extracellular Matrix,Invasion/Microenvironment,Metalloprotease,Proteolysis/Ubiquitin,Proteolytic Enzymes,Signal Transduction

We are currently using anti-BHC80/PHF21A antibody A08507-1 for human tissue, and we are happy with the ELISA results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on zebrafish tissues as well?
The anti-BHC80/PHF21A antibody (A08507-1) has not been validated for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in zebrafish you can get your next antibody for free. Please contact me if I can help you with anything.
