Product Overview

Clone AAEI-2 is a rabbit monoclonal antibody that recognizes BCL2/adenovirus E1B 19 kDa protein-interacting protein 3 (gene BNIP3) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 22.0 kDa; the supplier reports observed bands at 30, 50 kDa.

BNIP3 is a BH3-containing protein of the mitochondrial outer membrane that is strongly induced by hypoxia through HIF-1. It promotes mitochondrial autophagy (mitophagy) by binding LC3 family proteins and can also contribute to cell death, and it is studied in hypoxia, cardiac injury and cancer.

Samples with confirmed staining (IHC): human pancreatic cancer tissue, human placenta tissue, human thyroid cancer tissue, human kidney tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human BNIP3.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places BCL2/adenovirus E1B 19 kDa protein-interacting protein 3 in the mitochondrion and mitochondrion outer membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 22.0 kDa; observed bands at about 30 and 50 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human BNIP3.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
