Vendor description (summary): Boster Bio Anti-CBX1/HP1 Antibody Picoband® catalog # A04206-2.

Biological background

Biological context: Likely involved in the processes that promote cell division prior to the formation of differentiated tissues.

Expression and localization notes: cellular localization: Cytoplasm . Nucleus . Cytoplasm, cytoskeleton, microtubule organizing center, centrosome . Midbody . Nucleus during interphase. Weakly concentrated at centrosomes during mitosis and colocalizes with AURKC at the midbody during cytokinesis. ., tissue context: Isoform 1, isoform 3 and isoform 5 are ubiquitous. Isoform 2 is strongly expressed in the brain, weakly detectable in lung and colon, and overexpressed in gastric cancer. Isoform 4 is not detected in normal tissues, but strong expression was found in gastric cancer tissues. Down-regulated in a subset of cases of breast cancer. ..

Common research applications

  • Western blotting (WB): Compare CBX1 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of CBX1 in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify CBX1-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Background: Chromobox protein homolog 1 is a protein that in humans is encoded by the CBX1 gene. This gene encodes a highly conserved nonhistone protein, which is a member of the heterochromatin protein family . The protein is enriched in the heterochromatin and associated with centromeres. The protein has a single N-terminal chromodomain which can bind to histone proteins via methylated lysine residues, and a C-terminal chromo shadow-domain (CSD) which is responsible for the homodimerization and interaction with a number of chromatin-associated nonhistone proteins. The protein may play an important role in the epigenetic control of chromatin structure and gene expression. Several related pseudogenes are located on chromosomes 1, 3, and X. Multiple alternatively spliced variants, encoding the same protein, have been identified.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cytoplasm . Nucleus . Cytoplasm, cytoskeleton, microtubule organizing center, centrosome . Midbody . Nucleus during interphase. Weakly concentrated at centrosomes during mitosis and colocalizes with AURKC at the midbody during cytokinesis. .
  • Tissue details: Isoform 1, isoform 3 and isoform 5 are ubiquitous. Isoform 2 is strongly expressed in the brain, weakly detectable in lung and colon, and overexpressed in gastric cancer. Isoform 4 is not detected in normal tissues, but strong expression was found in gastric cancer tissues. Down-regulated in a subset of cases of breast cancer. .
