Overview

This antibody is intended for detection of ITGAM (Integrin alpha-M) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-CD11b/ITGAM Antibody Picoband® catalog # A00144-1. Tested in ELISA, Flow Cytometry, IHC, WB applications. This antibody reacts with Human, Mouse. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E. coli-derived human CD11b recombinant protein (Position: N129-E336). (reported region: N129-E336).
  • Molecular weight context: reported MW: 179 kDa; calculated MW: 127 kDa
  • Reactivity: Human,Mouse
  • Applications: ELISA, Flow Cytometry, IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Integrin alpha-M; integrin alpha M. Integrin alpha M (ITGAM) is one protein subunit that forms the heterodimeric integrin alpha-M beta-2 (αMβ2) molecule, also known as macrophage-1 antigen (Mac-1) or cluster of differentiation molecule 11B (CD11B). It is mapped to 16p11.2. ITGAM has a role in vascular repair after mechanical arterial injury. It is implicated in various adhesive interactions of monocytes, macrophages and granulocytes as well as in mediating the uptake of complement-coated particles. What's more, ITGAM probably recognizes the R-G-D peptide in C3b, and it is also a receptor for fibrinogen, factor X and ICAM1. Functional note: Integrin alpha-M/beta-2 is implicated in various adhesive interactions of monocytes, macrophages and granulocytes as well as in mediating the uptake of complement-coated particles. It is identical with CR-3, the receptor for the iC3b fragment of the third complement component. It probably recognizes the R-G-D peptide in C3b. Integrin alpha-M/beta-2 is also a receptor for fibrinogen, factor X and ICAM1. It recognizes P1 and P2 peptides of fibrinogen gamma chain. Reported localization: Membrane; Single-pass type I membrane protein. Expression/tissue context: Predominantly expressed in monocytes and granulocytes.

Research relevance and current trends

  • Cell Adhesion: Researchers commonly examine how ITGAM (Integrin alpha-M) relates to this theme using model systems and orthogonal readouts.
  • Cell Type Marker: Researchers commonly examine how ITGAM (Integrin alpha-M) relates to this theme using model systems and orthogonal readouts.
  • Cell Type Markers: Researchers commonly examine how ITGAM (Integrin alpha-M) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative ITGAM (Integrin alpha-M) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of ITGAM (Integrin alpha-M) across tissue regions and cell types using matched controls.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.
  • ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

I was wanting to use your anti-CD11b/ITGAM antibody for ELISA for rat bone marrow cell on frozen tissues, but I want to know if it has been tested for this particular application. Has this antibody been tested and is this antibody a good choice for rat bone marrow cell identification?
It shows on the product datasheet, A00144-1 anti-CD11b/ITGAM antibody has been validated for ELISA, Flow Cytometry, IHC, ICC, WB on human, mouse, rat tissues. We have an innovator award program that if you test this antibody and show it works in rat bone marrow cell in IHC-frozen, you can get your next antibody for free.
My team were satisfied with the WB result of your anti-CD11b/ITGAM antibody. However we have seen positive staining in liver cell membrane using this antibody. Is that expected? Could you tell me where is ITGAM supposed to be expressed?
From what I have seen in literature, liver does express ITGAM. Generally ITGAM expresses in cell membrane. Regarding which tissues have ITGAM expression, here are a few articles citing expression in various tissues: Blood, Pubmed ID: 1346576, 3539202 Liver, Pubmed ID: 19159218
