Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Mouse Monoclonal Mouse IgG1
  • Clone number: Clone: 5F8
  • Immunogen / epitope context: E. coli-derived human CD2AP recombinant protein (Position: K253-K337). (reported region: K253-K337).
  • Molecular weight context: reported MW: 80 kDa; calculated MW: nan
  • Reactivity: Human,Mouse,Rat
  • Applications: Flow Cytometry, IF, IHC, ICC, WB

As a monoclonal antibody, the reagent targets a defined epitope, supporting consistency across experiments; epitope masking by PTMs or conformational changes can affect signal.

Biological background

activating transcription factor 1; CD2 associated protein. CD2-associated protein is a protein that in humans is encoded by the CD2AP gene. This gene encodes a scaffolding molecule that regulates the actin cytoskeleton. The protein ly interacts with filamentous actin and a variety of cell membrane proteins through multiple actin binding sites, SH3 domains, and a proline-rich region containing binding sites for SH3 domains. The cytoplasmic protein localizes to membrane ruffles, lipid rafts, and the leading edges of cells. It is implicated in dynamic actin remodeling and membrane trafficking that occurs during receptor endocytosis and cytokinesis. Haploinsufficiency of this gene is implicated in susceptibility to glomerular disease. Functional note: Seems to act as an adapter protein between membrane proteins and the actin cytoskeleton. In collaboration with CBLC, modulates the rate of RET turnover and may act as regulatory checkpoint that limits the potency of GDNF on neuronal survival. Controls CBLC function, converting it from an inhibitor to a promoter of RET degradation. May play a role in receptor clustering and cytoskeletal polarity in the junction between T- cell and antigen-presenting cell. May anchor the podocyte slit diaphragm to the actin cytoskeleton in renal glomerolus. Also required for cytokinesis. Reported localization: Cytoplasm, cytoskeleton . Cell projection, ruffle . Colocalizes with F-actin and BCAR1/p130Cas in membrane ruffles. Located at podocyte slit diaphragm between podocyte foot processes (By similarity). During late anaphase and telophase, concentrates in the vicinity of the midzone microtubules and in the midbody in late telophase. Expression/tissue context: Widely expressed in fetal and adult tissues.

Research relevance and current trends

  • Actin Binding Proteins: Researchers commonly examine how CD2AP (activating transcription factor 1) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative CD2AP (activating transcription factor 1) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of CD2AP (activating transcription factor 1) across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
