Overview
This antibody is intended for detection of CLDN5 (Claudin-5) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.
Vendor notes: Boster Bio Anti-Claudin 5/CLDN5 Antibody catalog # RP1043. Tested in WB applications. This antibody reacts with Human, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.
Key elements and design rationale
- Antibody format: Rabbit Polyclonal Rabbit IgG
- Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human Claudin 5, different from the related mouse sequence by three amino acids and from the related rat sequence by four amino acids.
- Molecular weight context: reported MW: 23 kDa; calculated MW: 23147 MW
- Reactivity: Human,Rat
- Applications: WB
As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.
Biological background
Claudin-5; Claudin-5. Claudin-5 (CLDN5) also known as BEC1, is a protein that in humans is encoded by the CLDN5 gene. By use of yeast artificial chromosomes, Claudin-5 is localized between polymorphic markers D22S944 and D22S941 on chromosome 22q11, both of which are deleted in more than 80% of VCFS patients. This gene encodes a member of the claudin family. Claudins are integral membrane proteins and components of tight junction strands. Tight junction strands serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets. What's more, it plays a major role in tight junction-specific obliteration of the intercellular space. Functional note: Plays a major role in tight junction-specific obliteration of the intercellular space. . Reported localization: Cell junction, tight junction. Cell membrane; Multi-pass membrane protein.
Research relevance and current trends
- Cell Adhesion: Researchers commonly examine how CLDN5 (Claudin-5) relates to this theme using model systems and orthogonal readouts.
- Cytoskeleton/ECM: Researchers commonly examine how CLDN5 (Claudin-5) relates to this theme using model systems and orthogonal readouts.
- Signal Transduction: Researchers commonly examine how CLDN5 (Claudin-5) relates to this theme using model systems and orthogonal readouts.
Common research applications
- Western blotting: compare relative CLDN5 (Claudin-5) levels across conditions; band patterns may reflect isoforms and processing.
Notes for experimental interpretation
- Specificity notes: No cross reactivity with other proteins.
- Cross-reactivity: No cross-reactivity with other proteins
- Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
- Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
