Key elements and design rationale
- Antibody format: Rabbit Polyclonal Rabbit IgG
- Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human Connexin 45/GJA7, different from the related mouse and rat sequences by three amino acids.
- Molecular weight context: reported MW: 45 kDa; calculated MW: 45470 MW
- Reactivity: Human,Rat
- Applications: WB
As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.
Biological background
Gap junction gamma-1 protein; Gap junction gamma-1 protein. Gap junction gamma-1 protein (GJC1), also known as gap junction alpha-7 protein (GJA7) or connexin 45 (Cx45), is a protein that in humans is encoded by the GJC1 gene. The International Radiation Hybrid Mapping Consortium mapped the GJA7 gene to chromosome 17q21.31. This gene is a member of the connexin gene family. The encoded protein is a component of gap junctions, which are composed of arrays of intercellular channels that provide a route for the diffusion of low molecular weight materials from cell to cell. Functional note: One gap junction consists of a cluster of closely packed pairs of transmembrane channels, the connexons, through which materials of low MW diffuse from one cell to a neighboring cell. Reported localization: Cell membrane; Multi-pass membrane protein. Cell junction, gap junction. Expression/tissue context: In skin, expressed in epidermis and epidermal appendages but not in dermis. Expressed in all layers of the epidermis except the basal layer. In hair follicles, expressed in the suprabasal layer but not the basal layer. Also expressed in melanoma and carcinoma cell lines, fibroblasts and foreskin melanocytes.
Research relevance and current trends
- Cell Adhesion: Researchers commonly examine how GJC1 (Gap junction gamma-1 protein) relates to this theme using model systems and orthogonal readouts.
- Cytoskeleton/ECM: Researchers commonly examine how GJC1 (Gap junction gamma-1 protein) relates to this theme using model systems and orthogonal readouts.
- Signal Transduction: Researchers commonly examine how GJC1 (Gap junction gamma-1 protein) relates to this theme using model systems and orthogonal readouts.
Common research applications
- Western blotting: compare relative GJC1 (Gap junction gamma-1 protein) levels across conditions; band patterns may reflect isoforms and processing.
Notes for experimental interpretation
- Specificity notes: No cross reactivity with other proteins.
- Cross-reactivity: No cross-reactivity with other proteins
- Family / similarity context: Contains 1 HMG box DNA-binding domain.
- Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
- Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
